TPHA Microtitre Plate Kit

The TPHA Microtitre Plate Kit is a highly sensitive and reliable serological assay specifically designed for the detection of antibodies against Treponema pallidum, the causative agent of syphilis. This kit provides a complete solution for both clinical and research laboratories, ensuring accurate and reproducible results in routine serodiagnosis.

The assay is based on the use of TPHA Test Cells (R1), which are preserved avian erythrocytes coated with the pathogenic T. pallidum antigen (Nichols strain). During testing, the presence of antibodies in patient serum causes visible hemagglutination of the coated erythrocytes. To ensure specificity, non-specific reactions are detected and controlled using Control Cells (R2), which are avian erythrocytes without T. pallidum antigen. Residual non-specific reactions can further be eliminated through an absorption step using the Control Cells.

To enhance specificity against pathogenic T. pallidum, antibodies reacting with non-pathogenic treponemes are removed by incorporating Treponema Reiter extract in the cell suspension. All reagents are supplied at optimized concentrations and ready-to-use dilutions, which simplifies assay preparation and ensures consistent performance across all recommended testing methods.

The kit includes positive and negative controls, providing built-in quality checks for accurate result interpretation. It is suitable for serum samples and can be applied to all standard TPHA testing formats, making it a versatile tool for syphilis serodiagnosis in diverse laboratory settings.

Storage and Handling:

Reagents should be stored at 2–8°C and must not be frozen. Vials should be kept upright to prevent cell aggregation. If aggregates form, gently mix the vial before use to disperse the cells evenly. The shelf life of all components is 18 months under proper storage conditions.

Test Principle:

The TPHA assay is based on passive hemagglutination. Antibodies against T. pallidum in the patient’s serum bind to antigen-coated erythrocytes, resulting in visible agglutination.

Non-Specific Reaction Control

Control Cells (R2) detect and help eliminate non-specific agglutination. Residual non-specific reactions can be removed by an absorption step using Control Cells.

Specificity Enhancement

Antibodies against non-pathogenic treponemes are removed using Treponema Reiter extract present in the test cell suspension.

Sample Type

The assay is designed for use with human serum samples.

Reagent Preparation

All reagents are supplied at optimal dilutions and ready-to-use concentrations. No further dilution is required.

Kit Components

Includes TPHA Test Cells, Control Cells, TPHA Diluent, Positive Control, and Negative Control.

Storage Requirements

Store vials at 2–8°C. Do not freeze. Keep vials upright to avoid cell aggregation. Mix gently before use if aggregates form.

Shelf Life

All reagents remain stable for up to 18 months when stored correctly.

Applications

Suitable for clinical and research laboratories for syphilis serodiagnosis, quality control, and routine TPHA testing.
